Crispim,Bruno do Amaral; Seno,Leonardo de Oliveira; Egito,Andréa Alves do; Vargas Junior,Fernando Miranda de; Grisolia,Alexéia Barufatti.
Background The aim of the present study was to assess the genetic diversity of Pantaneiro sheep, using microsatellite markers, in order to assist maintenance and management plans, enhance mating systems and reduce the inbreeding rate. A total of 127 animals were genotyped at eight microsatellite loci. They belonged to populations from the Experimental Farm of the Universidade Federal da Grande Dourados (UFGD) (Dourados/MS/Brazil) and Embrapa Pantanal (Corumbá/MS/Brazil). Results The population of Pantaneiro sheep from the UFGD exhibited a high mean number of alleles (11.13) and allelic richness (10.66). The polymorphic information content was highly informative in the locus studied, resulting in a mean value of 0.71. Observed heterozygosity was lower than...

Bovine landscape genetics in Brazil Scientia Agricola
Souza,Ana Clara Barbosa de; Egito,Andréa Alves do; Peripolli,Vanessa; McManus,Concepta Margaret.
ABSTRACT The aim of this study was to explore spatial genetic structure patterns in cattle breeds adapted to local conditions in Brazil. We georeferenced 876 animals of ten breeds raised in Brazil kept in the Genebank of Brazilian Agricultural Research Corporation (Embrapa) by sample locations using the QGIS 2.4.0 software. The Mantel tests, spatial autocorrelation, and Monmonier tests were performed. The distances for spatial correlation tests ranged from 5 to 15 classes. The results indicated genetic discontinuities in cattle breeds from the Midwest, South, and Southeast of the country. Correlation between genetic and geographic distance was low, but significant. The Monmonier Maximum Distance Algorithm indicated an initial subdivision of Curraleiro and...

D-loop haplotype diversity in Brazilian horse breeds Genet. Mol. Biol.
Ianella,Patrícia; Albuquerque,Maria do Socorro Maués; Paiva,Samuel Rezende; Egito,Andréa Alves do; Almeida,Leonardo Daniel; Sereno,Fabiana T. P. S.; Carvalho,Luiz Felipe Ramos; Mariante,Arthur da Silva; McManus,Concepta Margaret.
Abstract The first horses were brought to Brazil by the colonizers after 1534. Over the centuries, these animals evolved and adapted to local environmental conditions usually unsuitable for exotic breeds, thereby originating locally adapted Brazilian breeds. The present work represents the first description of maternal genetic diversity in these horse breeds based on D-loop sequences. A D-Loop HSV-I fragment of 252 bp, from 141 horses belonging to ten Brazilian breeds / genetic groups (locally adapted and specialized breeds) were analysed. Thirty-five different haplotypes belonging to 18 haplogroups were identified with 33 polymorphic sites. Haplotype diversity (varying from 0.20 to 0.96) and nucleotide diversity (varying from 0.0039 to 0.0239) was lower...

Genetic characterization of the Moxotó goat breed using RAPD markers PAB
Oliveira,Rejane Rodrigues de; Egito,Andréa Alves do; Ribeiro,Maria Norma; Paiva,Samuel Rezende; Albuquerque,Maria do Socorro Maués; Castro,Silvia Ribeiro; Mariante,Arthur da Silva; Adrião,Manuel.
The objective of this study was to verify the genetic diversity between and within seven populations of Moxotó goat (n = 264) from the States of Pernambuco, Paraíba and Rio Grande do Norte, using RAPD (Random Amplified Polymorphic DNA). Moxotó, as well as other naturalized breeds, suffers genetic losses due to the indiscriminate miscegenation with breeds raised in the Northeast Region of Brazil. The genetic characterization of these genetic resources is essential to conservation and breeding programs. DNA was extracted from lymphocytes using a non-organic protocol. The 16 primers used were selected from 120 decamer oligonucleotide primers and generated 56 polymorphic bands. The analysis of molecular variance (AMOVA) showed that the greater part of total...

Genetic diversity and population structure of Brazilian native bovine breeds PAB
Serrano,Germana Maria; Egito,Andréa Alves do; McManus,Concepta; Mariante,Arthur da Silva.
Conservation and improvement strategies should be based on the association between genetic and phenotypic characteristics. The objective of this work was to characterize five native Brazilian cattle breeds (Caracu, Crioulo Lageano, Curraleiro, National Polled and Pantaneiro) and two commercial breeds (Holstein and Nellore) using RAPD technique to estimate genetic distances and variability between and within breeds. Genetic relationships were investigated using 22 primers which generated 122 polymorphic bands. Analysis of molecular variance indicated that most of the genetic variation lay among individuals within populations. The genetic variabilities between pairs of breeds were statistically significant. The smallest genetic divergence was between Crioulo...

Egito,Andréa Alves do; Fuck,Beatriz Helena; McManus,Concepta; Paiva,Samuel Rezende; Albuquerque,Maria do Socorro Maués; Santos,Sandra Aparecida; Abreu,Urbano Gomes Pinto de; Silva,Joaquim Augusto da; Sereno,Fabiana Tavares Pires de Souza; Mariante,Arthur da Silva.
Blood samples were collected from Pantaneiro Horses in five regions of Mato Grosso do Sul and Mato Grosso States. Arabian, Mangalarga Marchador and Thoroughbred were also included to estimate genetic distances and the existing variability among and within these breeds by RAPD-PCR (Random Amplified Polymorphic DNA - Polymerase Chain Reaction) molecular markers. From 146 primers, 13 were chosen for amplification and 44 polymorphic bands were generated. The analysis of molecular variance (AMOVA) indicated that the greatest portion of detected variability was due to differences between individuals within populations (75.47%). Analysis of the genetic variability between pairs of populations presented higher estimates for the five Pantaneiro populations with the...
